Sequencing and analysis of the complete mitochondrial genome of Nicrophorus maculifrons (Coleoptera:Silphidae)

    Nicrophorus maculifrons belongs to Nicrophorinae,Silphidae,Coleoptera. In this study,high-throughput sequencing technology was used to assemble the complete mitochondrial genome sequence of N .maculifrons . The study analyzed the sequence structure,base composition,codon usage frequency,and the secondary structure of tRNA genes of N .maculifrons . At the same time,two phylogenetic trees were constructed using Bayesian and maximum likelihood methods based on the sequences of 13 protein-coding genes of the N .maculifrons and seven other species of Silphidae. The results showed that the mitochondrial genome of N .maculifrons is 16692 bp,including 13 protein-coding genes,22 tRNAs,2 rRNAs and a control region. The genome exhibits a significant AT bias,with an A+T content of 76.42%. Among the 22 tRNA genes,all but four,which have partial structural deficiencies,exhibit the typical cloverleaf secondary structure. The results of two phylogenetic analyses based on 13 protein-coding genes showed that the phylogenetic relationship between the N .maculifrons and Nicrophorus nepalensis is the closest,which was consistent with the traditional morphological classification results.
