Advances in Recombinant Factor C Technology for Endotoxin Detection

    Bacterial endotoxins, the primary pyrogens, can induce fever and systemic inflammatory responses even at trace levels, and may progress to multiple organ dysfunction or death in severe cases. Consequently, developing highly sensitive and accurate endotoxin detection technologies is of critical importance. Although limulus amebocyte lysate (LAL) remains the gold standard for endotoxin detection, the overharvesting of horseshoe crabs whose blood is both scarce and irreplaceable has led to drastic population declines, exacerbating ecological risks and significantly con-straining reagent supply. Recombinant factor C (rFC), engineered via recombinant DNA technology, demonstrates superior performance metrics, including rapid detection kinetics, high sensitivity, strong specificity, and complete independence from animal-derived materials. As a strategic solution to the supply bottleneck of traditional LAL, rFC has been gradually recognized by pharmacopoeias worldwide and demonstrates broad application prospects.
