Super-Resolution Localization Microscopy with Scientific Complementary Metal Oxide Semiconductor Camera
Super-resolution localization microscopy (SRLM) is a representative super-resolution imaging technology. Low-light detector is an essential component in SRLM. Compared with traditional electron multiplying charge coupled device (EMCCD) cameras using serial output, scientific complementary metal oxide semiconductor (sCMOS) cameras with parallel output provide simultaneously large field of view, high imaging speed and low read noise, thus bring new opportunities for SRLM, especially on achieving video-rate imaging and large field of view imaging. At the same time, SRLM based on sCMOS camera is also facing several challenges which are mainly originated from the high-speed camera, including but not limited to massive data transmission, storage, and analysis. This paper introduces the principle and development of SRLM, and discusses the current status, opportunities and challenges of SRLM based on sCMOS camera.
